Abstract
Polysaccharides have been used as a a chemoimmunotherapy agent in clinic treatment of heart and blood diseases. The extraction conditions of polysaccharide from ultramicro-bacteria Glaciecola were studied. Effects of sample to water ratio, extracting temperature and extracting time on polysaccharide yield, mucosity and surface tension were observed in normal water extraction and ultrasonic treatment. The results showed that there were significant differences in polysaccharide yield and mucosity among groups of different extracting time(p<0.05). The optimum extraction conditions were as follows: 1∶15 ratio of sample to water (w:v), 100℃ extracting temperature and 4~6 h extracting period. Ultrasonic treatment was proved to be an efficient method to enhance the rate of polysaccharide yield and decrease the surface tension of solution(p<0.05). The yield by ultrasonic treatment for 2 h extracting period is the same as that by un-ultrasonic treatment for 6 h extracting period. 100 W ultrasonic power for 10~15 min and 4 h extracting period would be suggested. The polysaccharide yield from ultramicro-bacteria Glaciecola can reach 8.82%.
