Isolation of total glycosides and flavonoids from Eucommia ulmoides leaf by macroporous resin

Abstract
Technology for the isolation of total glycosides and flavonides from the leaves of Eucommia ulmoides by 9 kinds of macroporous resins was studied. Results show that the optimum resins are XDA-1 and X-5. The technical conditions for the separation are as follows: pH value of the sample solution loaded: 6, static absorption time: above 10 h, suitable desorption solvent to XDA-1: 40% ethanol for chlorogenic acid and aucubin, 80% ethanol for flavonoids; to X-5: 30% ethanol for chlorogenic acid and aucubin, and 70% ethanol for flavonids. The yield of total glycosides (crude product) extracted by using XDA-1 is 14.23%, in which the content of chlorogenic acid is 14.23%, of aucubin, 7.69%. The yield of flavonoids (crude product) extracted by using XDA-1 is 4.76% with the content of 15.82%. The yield of total glycosides (crude product) extracted by using X-5 is 7.35%, in which the content of chlorogenic acid is 15.39%, of aucubin, 9.07%. The yield of flavonoids (crude product) extracted by using X-5 is 5.11% with the content of 16.15%.
