Abstract
Method was introduced to refine phosphatidylcholine from rude soybean lecithin using alcohol and remove impurity by refrigerating crystal in the condition of low temperature. The related factors influencing the purity of phosphatidylcholine were studied through single-factor experiments. The response surface experiments were used to optimize the conditions of refinement. Results show that freezing time, temperature and the ratio of liquor to material have some influence on the purity of phosphatidylcholine. The significant factors are cooling time and freezing temperature. The optimization technology are obtained using the method of response surface experiments, the freezing time is 23.56 h, the freezing temperature is -20℃ and the ratio of liquor to material is 8.73∶1(V/m). In these conditions, the prediction purity of phosphatidylcholine is 70.73%. Through the verification test and High Performance Liquid Chromatography(HPLC) detection. The purity of phosphatidylcholine is 70.1% and the yield is 23.0%. Semi-works production confirmed the purity of phosphatidylcholine is 68.8% and the yield is 22.7%. All the works prove this method can be used for industrial manufacture.
